Cagrilintide, also known as AM833 or NN0174-0833, is a synthetic, acylated peptide analogue of amyline (37 amino acids) developed as a long-acting agonist of amylin receptors (AMYR) and calcitonin receptors (CTR). Within peptide science, Cagrilintide is studied for its interaction with AMYR/CTR receptor complexes, downstream signaling cascades, and long-term receptor-binding kinetics in preclinical research models (Lau D.C.W. et al., 2021).
History of Cagrilintide
Cagrilintide evolved from research into long-active amyline analogues. Natural amyline has a short plasma half-life, which made preclinical characterization difficult. Through amino acid substitutions and lipid acylation – similar to the optimization strategy applied in GLP-1 analogues – Cagrilintide has been designed as a more stable molecule with extended receptor interaction duration. This strategy represents an extension of peptide engineering within endocrine-focused research (Kruse T. et al., 2021).
